Photochemical decomposition of persistent perfluorocarboxylic acids (PFCAs) in water by use of persulfate ion (S2O8(2-)) was examined to develop a technique to neutralize stationary sources of PFCAs. Photolysis of S2O8(2-) produced highly oxidative sulfate radical anions (SO4-), which efficiently decomposed perfluorooctanoic acid (PFOA) and other PFCAs bearing C4-C8 perfluoroalkyl groups. The major products were F- and CO2; also, small amounts of PFCAs with shorter than initial chain lengths were detected in the reaction solution. PFOA at a concentration of 1.35 mM (typical of that in untreated wastewater after an emulsifying process in fluoropolymer manufacture) was completely decomposed by a photochemical system with 50 mM S2O8(2-) and 4 h of irradiation from a 200-W xenon-mercury lamp. The initial PFOA decomposition rate was 11 times higherthan with photolysis alone. All sulfur-containing species in the reaction solution were eventually transformed to sulfate ions by this method. This method was successfully applied to the decomposition of perfluorononanoic acid contained in a floor wax solution.  相似文献

Gene expression profiles in normal human gingival and dermal fibroblasts were investigated using DNA microarrays. Their fundamental characteristics were almost identical, but 5% of their genes were uniquely expressed. These results help us to choose an optimal cell source for effective fibroblast-based cell therapy that is dependent on differential gene expression profiles.  相似文献

In membrane bioreactors (MBRs) for wastewater treatment, membrane fouling, particularly biofouling caused by soluble microbial products (SMP), is a nuisance problem causing decreases in permeation flux. In a previous study, we identified primary biofoulants of microfiltration (MF) membranes in SMP as polysaccharides containing uronic acids that undergo inter- and intramolecular ionic cross-linking by polyvalent cations, forming a gelatinous mass that clogs membrane pores. In the present study, we therefore attempted to isolate biofoulant-degrading microorganisms from activated sludge on a polygalacturonic acid-overlaid agar medium and evaluate their efficiency for preventing biofouling of MF membranes. Among the isolates, the fungal strain HO1 identified as Phialemonium curvatum degraded 30% of polysaccharides containing uronic acids into smaller molecules in a SMP solution containing a high concentration of saccharides after 30 days of cultivation. Microfiltration tests using a laboratory-scale submerged MBR indicated that the filtration resistance of this degraded SMP solution was lower than that of the control SMP solution without fungal inoculation. Importantly, accumulation of gelatinous mass on the membrane responsible for biofouling was avoided in the SMP solution augmented with P. curvatum HO1 during the microfiltration test. This is the first report to describe a new method for avoiding biofouling of MBRs by microbial degradation of primary biofoulants.  相似文献

Horseradish peroxidase isozyme C1a (HRP C1a) is widely used as a reporter enzyme in a variety of detection procedures such as enzyme-linked immunosorbent assay (ELISA) and western blotting. We previously isolated the gene encoding HRP C1a and showed that HRP C1a is at first translated as a preproprotein containing propeptides at its N- and C-termini (N-terminal secretion signal peptide and C-terminal propeptide; CTPP). The signal peptide (sp) is necessary for endoplasmic reticulum (ER) translocation and the CTPP acts as a vacuolar sorting determinant. Furthermore, HRP C1a was secreted into the culture medium from cells expressing the HRP C1a gene without the CTPP region. We optimized the secretory production system of HRP C1a in tobacco plants. To determine a suitable signal peptide for high-efficient secretion of proteins, three types of sp derived from HRP C1a (C1Psp), beta-D-glucan exohydrolase (GEsp) and 38 kDa peroxidase (38Psp) were compared. GE and 38P are secretory proteins highly accumulated in the culture medium of BY2 cells. The secretion efficiency was increased by 34% and 53% when GEsp and 38Psp was used, respectively. Next, we used a translational enhancer, the 5'-untranslated region of Nicotiana tabacum alcohol dehydrogenase gene (NtADH 5'-UTR). The production of HRP C1a was increased by placing NtADH 5'UTR in front of the ORF in BY2 cells. These results indicate that the localization and expression level of recombinant proteins can be controlled by the use of propeptides and 5'UTR, respectively. Finally, high-efficiency secretory production of the HRP C1a was also achieved in transgenic tobacco.  相似文献

Decomposition of perfluorooctanesulfonate (PFOS) and related chemicals in subcritical water was investigated. Although PFOS demonstrated little reactivity in pure subcritical water, addition of zerovalent metals to the reaction system enhanced the PFOS decomposition to form F-ions, with an increasing order of activity of no metal approximately equal Al < Cu < Zn < Fe. Use of iron led to the most efficient PFOS decomposition: When iron powder was added to an aqueous solution of PFOS (93-372 microM) and the mixture was heated at 350 degrees C for 6 h, PFOS concentration in the reaction solution fell below 2.2 microM (detection limit of HPLC with conductometric detection), with formation of F-ions with yields [i.e., (moles of F- formed)/(moles of fluorine content in initial PFOS) x 100] of 46.2-51.4% and without any formation of perfluorocarboxylic acids. A small amount of CHF3 was detected in the gas phase with a yield [i.e., (moles of CHF3)/(moles of carbon content in initial PFOS) x 100] of 0.7%, after the reaction of PFOS (372 microM) with iron at 350 degree C for 6 h. Spectroscopic measurements indicated that PFOS in water markedly adsorbed on the iron surface even at room temperature, and the adsorbed fluorinated species on the iron surface decomposed with rising temperature, with prominent release of F- ions to the solution phase above 250 degrees C. This method was also effective in decomposing other perfluoroalkylsulfonates bearing shorter chain (C2-C6) perfluoroalkyl groups and was successfully applied to the decomposition of PFOS contained in an antireflective coating agent used in semiconductor manufacturing.  相似文献

Degradation of C4F9C2H4OH in air over TiO2 particles was examined in this first report of gas-solid heterogeneous photochemical degradation of fluorotelomer alcohols (FTOHs), which may be precursors of perfluorocarboxylic acids (PFCAs) in the environment. Photoirradiation (>290 nm) of C4F9C2H4OH in air flowing over TiO2 produced CO2, via C4F9CH2CHO, C4F9CHO, CnF(2n+1)COF (n=2 and/or 3), and COF2, in that order. X-ray photoelectron spectroscopy of the Ti02 surface showed a decrease in the amount of fluorine bonded to carbon and an increase in the amount of F- as the degradation of C4F9C2H4OH in air proceeded. Of the carbon content in the initial C4F9C2H4OH (78.8 ppmv), 90.7% was transformed to CO2, and the predominant fluorine species produced on the TiO2 surface was F-. Fluorotelomer unsaturated acids, which are considered to be toxic and have been observed in the biodegradation of FTOHs, did notform. Increased relative humidity in the air accelerated the decomposition of the reaction intermediates, which led to increased CO2 and F- formation. This result indicates that humidity is a key factor for counteracting FTOHs in indoor air. Although perfluoroalkyl substances such as PFCAs in water reportedly undergo little photodegradation over TiO2, our data show that mineralization of C4F9C2H4OH in air can be achieved with TiO2.  相似文献

In this paper, a novel perfect tracking control method based on multirate feedforward control is proposed. The advantages of the proposed method are that: (1) the proposed multirate feedforward controller eliminates the notorious unstable zero problem in designing the discrete-time inverse system; (2) the states of the plant match the desired trajectories at every sampling point of reference input; and (3) the proposed controller is completely independent of the feedback characteristics. Thus, highly robust performance is assured by the robust feedback controller. Moreover, by generalizing the relationship between the sampling period of plant output and the control period of plant input, the proposed method can be applied to various systems with hardware restrictions of these periods, which leads to higher performance. Next, it is shown that the structure of the proposed perfect tracking controller is very simple and clear. Illustrative examples of position control using a DC servomotor are presented, and simulations and experiments demonstrate the advantages of this approach  相似文献

In order to understand the dyeing behaviour of synthetic fibres in supercritical carbon dioxide, the solubility of some disperse dyes in supercritical fluid, as well as the rate of dyeing and the equilibrium adsorption of these dyes, have been studied. Dye solubility was measured by a dynamic analytic method at a range of pressure (7.5–25 MPa) and temperature (50–145 °C). The apparent rate of dyeing was measured and the dyeing isotherm was obtained by plotting the equilibrium dye adsorption against the equilibrium dyebath concentration. Linear isotherms were obtained when poly(ethylene terephthalate) samples were dyed with the disperse dyes. The mechanism of dyeing using supercritical carbon dioxide was discussed by considering the solubility, the dyeing rate and the dyeing isotherm.  相似文献
